Feb 02 - Location of cities and buildings

Hello/Hola,

When describing the locations of cities or buildings, is it best to say "en" or "esta/estan"?

For instance, can I say:

"Mi novia y yo viven en Londres. Es una ciudad esta Inglaterra de Sur, y el Reino Unido."

or

"Mi esposa y yo son alumnos a una universidad en Nueva York. Esta universidad tiene una libraria buena en Manhattan."

I have missed out the accents, and I apologise for this as I am pushed for time currently.

tl;dr, can "en" or "está/están" be used interchangeably for locations?
